## Introduction The blood-brain barrier (BBB) represents one of the most formidable challenges in neurological drug delivery. Composed of tightly joined endothelial cells, this selective membrane protects the brain from systemic circulation — but also blocks more than 98% of small-molecule drugs and nearly all large-molecule therapeutics. At Nobil Laboratories, our formulation division has spent three years engineering lipid nanoparticles (LNPs) capable of receptor-mediated transcytosis across this barrier. The results from our preclinical programme are now ready for disclosure. ## What Are Lipid Nanoparticles? Lipid nanoparticles are nanoscale (50–200 nm) spherical vesicles composed of ionisable lipids, phospholipids, cholesterol, and PEG-lipids. Originally developed for mRNA vaccine delivery, their versatility has expanded dramatically. Our modified LNP formulation incorporates a transferrin-receptor targeting ligand on the outer leaflet, enabling active transport into the CNS. ## Key Findings from Preclinical Work In our murine model, modified LNPs achieved a 4.2-fold increase in CNS drug accumulation compared to free-drug controls. Crucially, systemic off-target distribution was reduced by 31%, suggesting improved therapeutic windows for future candidates. Encapsulation efficiency across three model compounds exceeded 89%, and particle stability was maintained at 4°C for 18 months — a critical requirement for commercial viability. ## Next Steps We are now preparing an Investigational New Drug (IND) application for our lead CNS candidate. A Phase I first-in-human study is targeted for Q3 2026, enrolling patients with early-stage Alzheimer's disease at two sites in India and the United Kingdom. ## Conclusion Lipid nanoparticle technology represents a paradigm shift in how we approach CNS drug delivery. The convergence of receptor targeting, scalable manufacturing, and proven safety profiles makes this one of the most promising platforms in our pipeline.
